Biological Background: OVOL2 Function and Localization
- OVOL2 functions as a zinc-finger transcription repressor. Related references: PMID:19700410
- It suppresses keratinocyte cell cycling and terminal differentiation by directly repressing MYC and NOTCH1. Related references: PMID:19700410
- OVOL2 maintains epithelial identity by suppressing epithelial-to-mesenchymal transition (EMT) through repression of ZEB1.
- It positively regulates neuronal differentiation and is important for the correct development of primordial germ cells in embryos.
- OVOL2 plays dual roles in thermogenesis and adipogenesis to maintain energy balance, essential for brown/beige adipose tissue-mediated thermogenesis and limiting adipogenesis by blocking CEBPA activity.
- OVOL2 is expressed in testis, ovary, heart, and skeletal muscle. Related references: PMID:12213202
- It is expressed in the cornea but absent from the corneal endothelium, and mutations are linked to corneal dystrophy. Related references: PMID:26749309
- The protein localizes to the nucleus, contains zinc-finger domains, and undergoes alternative splicing and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen encompasses the N-terminal region (aa 24–98) of human OVOL2; this sequence may be conserved in mouse and rat based on cross-reactivity data.
- For Western blotting, consider using nuclear extracts from testis, ovary, heart, or skeletal muscle where OVOL2 is expressed.
- For ELISA applications, validate coating conditions and detection range with the recombinant immunogen as a positive control.
- Cross-reactivity with mouse and rat has been noted, but independent validation in the relevant experimental system is recommended.
- As a polyclonal antibody, batch-to-batch consistency should be assessed for quantitative assays.
