Biological Background: OTUD7B Function and Localization

  • OTUD7B (Cezanne, ZA20D1) is an OTU domain-containing deubiquitinase that negatively regulates the non-canonical NF-κB pathway by deubiquitinating TRAF3, preventing its proteolysis and subsequent pathway activation. Related references: PMID:18178551
  • In vitro, OTUD7B deubiquitinates 'Lys-11', 'Lys-48', and 'Lys-63'-linked polyubiquitin chains with a preference for 'Lys-11' linkages; it also hydrolyzes linear and branched polyubiquitin. However, the physiological relevance of these specificities is uncertain. Related references: PMID:11463333, PMID:20622874, PMID:23827681, PMID:27732584, PMID:12682062
  • OTUD7B deubiquitinates EGFR and ZAP70, modulating growth factor signaling and T cell receptor signaling, respectively, which impacts T cell homeostasis and NF-κB activation. Related references: PMID:22179831, PMID:26903241
  • It acts as a regulator of mTOR complex assembly by deubiquitinating MLST8, promoting mTORC2 formation while inhibiting mTORC1, thus influencing cell growth and metabolism. Related references: PMID:28489822
  • Localizes to both the cytoplasm and nucleus, indicating diverse signaling roles.
  • Widely expressed with high levels in kidney, heart, and fetal liver; differentially expressed during B-cell development, higher in immature B-cells than mature ones.
  • Contains a zinc finger domain and belongs to the A20-type deubiquitinase family; classified as a thiol protease involved in the ubiquitin conjugation pathway and immunity.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 530-700, covering the C-terminal region that may include the zinc finger domain; antibodies raised against this region are likely to detect full-length OTUD7B.
  • For Western blot (WB), expect a band at ~93 kDa; use appropriate positive controls such as HEK293 or Jurkat cell lysates.
  • For immunohistochemistry (IHC-P), antigen retrieval may be required due to the nuclear/cytoplasmic localization; validate antibody specificity using tissue sections with known expression patterns.
  • Since OTUD7B is conserved across human, mouse, and rat, the antibody can be used for cross-species studies, but confirm reactivity in the relevant sample system.
