Biological Background: OTUD4 Function and Localization

  • OTUD4, also known as HIV-1-induced protein HIN-1, is a deubiquitinase that hydrolyzes the isopeptide bond between ubiquitin and lysine residues on target proteins (PubMed:23827681, PubMed:25944111, PubMed:29395066).
    Related references: PMID:23827681 PMID:25944111 PMID:29395066
  • Upon phosphorylation at Ser-202 and Ser-204, OTUD4 specifically deubiquitinates Lys-63-polyubiquitinated MYD88 adapter protein via IL-1 receptor and Toll-like receptor pathways, attenuating NF-kB-dependent inflammatory transcription (PubMed:29395066).
    Related references: PMID:29395066
  • Independently of its catalytic activity, OTUD4 serves as a scaffold to assemble USP7 and USP9X deubiquitinases, stabilizing the alkylation repair enzyme ALKBH3 and promoting repair of alkylated DNA lesions (PubMed:25944111).
    Related references: PMID:25944111
  • The protein localizes to both the cytoplasm and nucleus, consistent with its roles in immune signaling and DNA repair.
  • OTUD4 undergoes post-translational modifications including phosphorylation (at least at Ser-202/Ser-204) and acetylation, and is subject to alternative splicing.
  • The full-length protein has a calculated molecular weight of approximately 124 kDa (UniProt Q01804).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 200-500 of human OTUD4, a central region that may detect endogenous OTUD4 in multiple application formats.
  • For Western blotting, the full-length OTUD4 protein runs at approximately 124 kDa; a 6-8% SDS-PAGE gel and appropriate transfer conditions are recommended.
  • For immunofluorescence/ICC, OTUD4 exhibits both cytoplasmic and nuclear localization; permeabilization conditions should be optimized to detect nuclear pools.
  • ELISA experiments may be performed using the recombinant immunogen or cell lysates; consider validating with a known positive control.
  • Cross-reactivity is expected with human, mouse, and rat OTUD4 based on sequence homology; however, performance in each species should be confirmed experimentally.
  • As a polyclonal antibody, lot-to-lot consistency should be verified, and it is advisable to include appropriate positive and negative controls in all assays.
