  • Otoferlin (Fer-1-like protein 2) is a multi-isoform protein; isoform 1 and 3 are expressed in adult brain, while isoform 2 is found in fetus and adult brain, heart, placenta, skeletal muscle, and kidney.
  • Functions as a key calcium sensor at ribbon synapses of cochlear inner hair cells (IHCs), triggering synaptic vesicle exocytosis and neurotransmitter release by interacting with presynaptic SNARE proteins in a calcium-dependent manner.
  • Also essential for synaptic exocytosis in immature outer hair cells (OHCs) and may play a role in endosome recycling.
  • Belongs to the ferlin family of proteins, characterized by multiple C2 domains that mediate calcium-dependent membrane interactions.
  • Subcellularly localizes to the cytoplasmic vesicle, synaptic vesicle membrane, basolateral cell membrane, endoplasmic reticulum membrane, Golgi apparatus membrane, presynaptic cell membrane, cell membrane, and trans-Golgi network.
