Biological Background: Osteopontin Function and Localization
- Osteopontin (OPN), also known as bone sialoprotein 1, secreted phosphoprotein 1, and uropontin, is a major non-collagenous bone matrix protein encoded by the SPP1 gene.
- It binds tightly to hydroxyapatite and forms an integral part of the mineralized extracellular matrix, likely mediating cell-matrix interactions.
- OPN also acts as a cytokine, enhancing production of interferon-gamma and interleukin-12 while reducing interleukin-10, essential for type I immunity.

- Post-translational modifications such as glycosylation, phosphorylation, and sialylation contribute to its function; it exists as a glycoprotein, phosphoprotein, and a proteoglycan.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal to central region (aa 17-314) of human osteopontin, covering most of the mature protein sequence. Due to high sequence homology, this antibody may detect isoforms and other species variants.
- For Western blot, osteopontin has a predicted molecular weight of ~35 kDa, but post-translational modifications can cause shifts; validate with appropriate positive control lysates from bone or kidney tissue.
- For ELISA, consider using recombinant osteopontin as a standard, and test cross-reactivity with mouse and rat proteins.
- Since osteopontin is secreted, cell culture supernatants or biological fluids like serum/urine may be suitable samples, but optimize dilution and blocking conditions.
