Biological Background: Osteopontin Function and Localization

  • Osteopontin (OPN), also known as bone sialoprotein 1, secreted phosphoprotein 1, and uropontin, is a major non-collagenous bone matrix protein encoded by the SPP1 gene.
  • It binds tightly to hydroxyapatite and forms an integral part of the mineralized extracellular matrix, likely mediating cell-matrix interactions.
  • OPN also acts as a cytokine, enhancing production of interferon-gamma and interleukin-12 while reducing interleukin-10, essential for type I immunity.
  • The protein is secreted and found in body fluids; detected in cerebrospinal fluid, urine, and plasma, with high expression in bone. Related references: PMID:25326458 PMID:36213313 PMID:37453717
  • Post-translational modifications such as glycosylation, phosphorylation, and sialylation contribute to its function; it exists as a glycoprotein, phosphoprotein, and a proteoglycan.
  • Alternative splicing produces multiple isoforms, which may exhibit tissue-specific functions.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal to central region (aa 17-314) of human osteopontin, covering most of the mature protein sequence. Due to high sequence homology, this antibody may detect isoforms and other species variants.
  • For Western blot, osteopontin has a predicted molecular weight of ~35 kDa, but post-translational modifications can cause shifts; validate with appropriate positive control lysates from bone or kidney tissue.
  • For ELISA, consider using recombinant osteopontin as a standard, and test cross-reactivity with mouse and rat proteins.
  • Since osteopontin is secreted, cell culture supernatants or biological fluids like serum/urine may be suitable samples, but optimize dilution and blocking conditions.
