Biological Background: OsNPR1 Function and Localization
- OsNPR1 (NPR1 homolog 1, OsNH1) is a rice protein containing a BTB/POZ domain, ankyrin repeats, and a zinc-finger, functioning in plant defense signaling.
- Acts as a salicylic acid receptor and substrate adapter for the CUL3-RBX1 E3 ubiquitin ligase complex, targeting proteins for ubiquitin-dependent degradation.
- Functions as a transcription cofactor: represses gene expression via WRKY factors in the absence of SA, and activates expression via TGA factors when sumoylated and SA is present.
- Essential for benzothiadiazole (BTH)-induced resistance against rice blast fungus (Magnaporthe oryzae). Related references: PMID:20924648
- Contributes to defense against bacterial blight (Xanthomonas oryzae pv. oryzae); overexpression enhances disease resistance but increases herbivore susceptibility. Related references: PMID:17309686
- Exhibits transcriptional coactivator activity with TGA2.1 and LG2 in vitro. Related references: PMID:22353606
- Modulates herbivore-induced defense responses; silencing reduces trypsin proteinase inhibitors and volatiles, enhancing resistance to Chilo suppressalis. Related references: PMID:22694163
- Localizes to the cytoplasm, nucleus, and nuclear bodies, consistent with its roles in signaling and transcriptional regulation.
Experimental Guidance and Technical Tips
- The immunogen spans a region near the C-terminus (residues 475–582), which may be suitable for detecting full-length OsNPR1 in Western blotting under reducing conditions.
- For Western blot, consider using appropriate positive control lysates from rice (Oryza sativa) tissues known to express OsNPR1, such as leaves after pathogen or SA treatment.
- In ELISA, validate the antibody with the recombinant immunogen as a standard and test cross-reactivity with related NPR1-like proteins if needed.
- Since OsNPR1 undergoes sumoylation and phosphorylation, post-translational modifications may affect antibody recognition; verify in the relevant sample system.
