Biological Background: OSGIN1 Function and Localization

  • OSGIN1 (oxidative stress-induced growth inhibitor 1), also known as BDGI (bone marrow stromal cell-derived growth inhibitor), OKL38 (ovary, kidney and liver protein 38), or huOKL38, is a pregnancy-induced growth inhibitor.
  • Possesses monooxygenase catalytic activity and regulates cytokinesis by promoting RHOA activity, likely acting locally at the midbody during late cytokinesis. This activity stabilizes transient intercellular bridges between daughter cells before abscission. Related references: PMID:38442170
  • Regulates cell differentiation and proliferation through the control of cell death. Related references: PMID:11459809 PMID:14570898
  • Localizes to the midbody, a structure critical for the final stages of cell division.
  • Ubiquitously expressed, with highest levels in ovary, testis, kidney, skeletal muscle, and liver. Related references: PMID:11459809 PMID:14570898
  • Classified as a developmental protein and differentiation factor; associated with keywords FAD, Flavoprotein, Growth regulation, Monooxygenase, NADP, Oxidoreductase, and Phosphoprotein.
  • Contains a phosphoprotein modification, indicating potential regulation by phosphorylation.
  • The mature protein has a calculated molecular weight of approximately 52 kDa.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 1–200 of human OSGIN1, an N-terminal region; consider this when designing epitope-specific assays.
  • For Western blot, use reducing SDS-PAGE conditions and include a molecular weight marker to verify the predicted band near 52 kDa.
  • For ELISA, validate with the recombinant immunogen or cell lysates known to express OSGIN1; a standard curve using the recombinant protein is recommended.
  • The antibody is verified for human reactivity; cross-reactivity with other species should be validated individually in the relevant sample system.
