Biological Background: OsELF3 Function and Localization
- OsELF3, also known as HD3B, EF7, or ELF3-1, is a rice protein involved in the photoperiodic regulation of flowering time. It acts as a floral promoter by negatively regulating GHD7, a repressor of flowering under both short day (SD) and long day (LD) conditions. Related references: PMID:22399582 PMID:22422935 PMID:22912900
- It functions as a floral activator specifically within the LD photoperiodic pathway. Related references: PMID:22888152
- Under SD conditions, OsELF3 is involved in blue light-induced activation of EHD1 expression, promoting flowering. Related references: PMID:22912900
- The protein localizes to the nucleus, consistent with its role in transcriptional regulation.
- Tissue-specific expression is observed in mesophyll cells of young leaves, anthers, stigmas, and the top of lemmas.
- UniProt keywords highlight involvement in flowering, transcription regulation, nucleus localization, and inclusion in the reference proteome.
Experimental Guidance and Technical Tips
- This antibody was generated against an N‑terminal recombinant fragment (amino acids 1–172) of OsELF3. Consider this immunogen design when assessing isoform coverage or potential cross‑reactivity with other ELF3 family members.
- For Western blot (WB) detection, the predicted molecular weight of OsELF3 is approximately 82 kDa. Validate in the relevant rice sample system, and optimize loading controls and transfer conditions accordingly.
- In ELISA applications, the antibody may be used as a capture or detection reagent. Determine optimal coating concentrations and blocking conditions empirically for your assay format.
- Given the role of OsELF3 in flowering time regulation, consider expression profiling across developmental stages or photoperiodic conditions when designing experiments.
