Biological Background: OSBPL3 Function and Localization
- OSBPL3 (ORP3) is a member of the oxysterol-binding protein-related protein family, also annotated as a phosphoinositide transfer protein. Multiple isoforms (e.g., 1a-1d, 2a-2d) are generated by alternative splicing and exhibit tissue-specific expression patterns.
- The protein functions as a lipid transfer protein that mediates non-vesicular transport of phosphoinositide 4-phosphate (PI(4)P) at plasma membrane (PM)/endoplasmic reticulum (ER) contact sites, extracting PI4P from the PM in exchange for phosphatidylcholine (PubMed:32234213).

PMID:32234213 - OSBPL3 interacts with the ER membrane protein VAPA, recruiting it to PM sites and thereby linking ER and PM compartments (PubMed:25447204).
Related references:
PMID:25447204 - The ORP3-VAPA complex stimulates RRAS signaling, which attenuates integrin beta-1 (ITGB1) activation at the cell surface (PubMed:18270267, PubMed:25447204).

PMID:18270267
PMID:25447204 - Subcellular localization includes the endoplasmic reticulum membrane, cytoplasm, cytosol, cell membrane, filopodium tips, and nucleus membrane, consistent with its role at membrane contact sites.
- OSBPL3 binds phosphoinositides with preference for PI(3,4)P2 and PI(3,4,5)P3, as well as 25-hydroxycholesterol and cholesterol (PubMed:16143324, PubMed:17428193).

PMID:17428193 - The protein has been implicated in regulation of the actin cytoskeleton, cell polarity, and cell adhesion (PubMed:18270267).

PMID:18270267 - Tissue expression: highly expressed in kidney, lymph node, and thymus; moderately expressed in stomach, jejunum, ileum, appendix, spleen, leukocytes, trachea, lung, and thyroid; low in brain and other tissues (PubMed:14593528). Isoforms show differential tissue distribution (PubMed:12590732).

Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 370-520 of human OSBPL3. This region may contain functional domains relevant to lipid binding or protein interactions.
- Suitable for Western blot (WB) and ELISA applications. For WB, consider testing multiple tissue or cell lysates (e.g., kidney, lymph node) known to express OSBPL3.
- Cross-reactivity with mouse and rat OSBPL3 is listed; validate reactivity in the relevant model systems before use.
- As a polyclonal antibody, lot-to-lot consistency should be monitored, especially for quantitative assays.
- Due to the presence of multiple isoforms and potential post-translational modifications, observed band sizes may differ from the calculated molecular weight (101 kDa).
