Biological Background: OSBPL2 Function and Localization
- OSBPL2 (Oxysterol-binding protein-related protein 2), also known as ORP2, is an intracellular lipid transport protein that binds sterols and phospholipids, mediating lipid transport between intracellular compartments. Related references: PMID:30581148, PMID:32914503, PMID:34124795
- It increases plasma membrane cholesterol levels and decreases phosphatidylinositol-4,5-bisphosphate (PIP2) levels, controlling LDL-cholesterol plasma membrane delivery, focal adhesion kinase (FAK) activation, and PI(4,5)P2 generation, thereby enhancing cell adhesion. Related references: PMID:30581148, PMID:32914503, PMID:34124795
- Exhibits strong binding to phosphatidic acid and weak binding to phosphatidylinositol 3-phosphate; also binds cholesterol, dehydroergosterol, 22(R)-hydroxycholesterol, and 25-hydroxycholesterol in vitro. Related references: PMID:11279184, PMID:17428193, PMID:19224871, PMID:30581148
- In conjunction with PLCB3, OSBPL2 may contribute to the control of keratinocyte proliferation and differentiation, likely through regulation of the ERK pathway and cell cycle. Related references: PMID:38701954
- Involved in angiogenic signaling pathways such as VEGFR2 in endothelial cells. Related references: PMID:32914503
- Localizes to the cytoplasm, cytosol, lipid droplets, cell membrane, and stereocilia (cell projections).
- Widely expressed across tissues; associated with deafness (DFNA67) and ichthyosis. Keywords include alternative splicing, lipid transport, lipid-binding, and non-syndromic deafness.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a recombinant fragment covering amino acids 291-480 of human OSBPL2 (NP_653081.1), which may generate antibodies recognizing multiple epitopes within this region. Consider epitope mapping if specific domain recognition is required.
- For Western blot (WB), a band around 55 kDa is expected; however, validate the observed band size in your specific sample system, as post-translational modifications or isoforms may alter migration.
- ELISA can be used for titer determination or screening; ensure proper coating and detection conditions for the recombinant immunogen or native protein.
- Given the polyclonal nature, cross-reactivity with other OSBP-related proteins cannot be excluded; perform appropriate controls for your target species.
