Biological Background: ORP150/GRP170/HSP12A Function and Localization
- Hypoxia up-regulated protein 1 (HYOU1), also known as ORP-150, GRP-170, and heat shock protein family H member 4, is a molecular chaperone.
- It plays a pivotal role in cytoprotective mechanisms triggered by oxygen deprivation; promotes HSPA5/BiP-mediated ATP nucleotide exchange and activates the unfolded protein response (UPR) pathway under endoplasmic reticulum (ER) stress (By similarity).
- The protein localizes to the endoplasmic reticulum lumen.
- HYOU1 is highly expressed in tissues with extensive ER and high secretory protein synthesis, such as liver and pancreas; lower expression is found in brain and kidney. It is also present in macrophages within aortic atherosclerotic plaques and in breast cancers.
- The protein contains ATP-binding and nucleotide-binding domains, characteristic of heat shock proteins.
- Post-translational modifications include acetylation, glycosylation, and phosphorylation, as indicated by keywords.
- Alternative splicing and disease variants have been reported for this gene.
Experimental Guidance and Technical Tips
- For Western blot (WB), expect a band at approximately 111 kDa. Consider using positive control lysates from liver or pancreas, which express high levels of HYOU1.
- For immunohistochemistry (IHC-P), validate tissue fixation and antigen retrieval protocols, as the target is located in the ER lumen. Include positive tissue controls such as liver or atherosclerotic plaque sections.
- For ELISA, we recommend titrating the antibody as a capture or detection reagent and validating performance with recombinant protein or cell lysates.
- The immunogen spans the N-terminal region (aa 1-150), so the antibody may recognize variants containing this epitope. Validate specificity in your experimental system.
