Biological Background: ORP1 Function and Localization
- ORP1, encoded by the OSBPL1A gene, is an oxysterol-binding protein-related protein, also known as OSBP8 or OSBPL1B. It features ankyrin repeats and a coiled-coil domain, with a molecular weight of approximately 108 kDa.
- The protein specifically localizes to late endosomes, where it plays a critical role in lipid transport and endosomal function.
- ORP1 binds phospholipids, exhibiting strong affinity for phosphatidic acid and weaker affinity for phosphatidylinositol 3-phosphate (By similarity).

- For western blotting, expect a band at ~108 kDa. Detection should be validated in human, mouse, and rat lysates as per cross-reactivity data.
- For immunofluorescence or immunocytochemistry, optimize fixation and permeabilization conditions to effectively access late endosomal targets.
