Biological Background: OPTN Function and Localization
- Optineurin (OPTN), also known as FIP-2, HIP-7, or NRP, is a ~66 kDa multidomain protein containing zinc finger and coiled-coil motifs.
- Plays a critical role in Golgi maintenance and membrane trafficking by linking myosin VI to the Golgi, essential for Golgi ribbon formation. Related references: PMID:27534431
- Functions in innate immunity: recruits TBK1 to the Golgi upon viral infection, leading to IRF3 phosphorylation and IFN-beta production. Related references: PMID:27538435
- Acts as a selective autophagy receptor, targeting ubiquitinated bacteria (e.g., Salmonella) for degradation via LC3-interaction, cooperating with SQSTM1 and CALCOCO2/NDP52.

- The full-length immunogen (aa 1-577) detects endogenous OPTN across multiple assays. For Western blot, a predominant band around 66 kDa is expected; include appropriate positive and negative controls to confirm specificity.
- For immunohistochemistry (paraffin-embedded sections, IHC-P), consider optimizing antigen retrieval and signal detection; validate staining in tissues known to express OPTN, such as human skeletal muscle or mouse brain.
- This antibody is compatible with immunoprecipitation (IP) and ELISA; for IP, use a starting amount of 1-5 µg per 500 µg lysate and corroborate with target-specific co-IPs.
