Biological Background: OPRL1 Function and Localization
- OPRL1 encodes the nociceptin receptor, also known as kappa-type 3 opioid receptor (KOR-3) or orphanin FQ receptor, a member of the G protein-coupled receptor (GPCR) family.
- It serves as a receptor for the endogenous neuropeptide nociceptin/orphanin FQ, playing a key role in modulating pain perception and locomotor activity.
- Upon ligand binding, the receptor activates intracellular G proteins, leading to inhibition of adenylate cyclase and calcium channels, and can also signal through arrestin-mediated pathways to activate MAP kinases.
- The protein localizes primarily to the cell membrane and cytoplasmic vesicles, and tissue specificity has been demonstrated in blood leukocytes.
- Alternative splicing of OPRL1 produces multiple transcript variants, potentially yielding receptor isoforms with distinct functions.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 270–370 within the intracellular C-terminal region of human OPRL1. For Western blot (WB), consider using denaturing and reducing conditions to detect the predicted ~41 kDa band.
- For ELISA applications, the antibody may recognize the linear peptide epitope; ensure appropriate coating and blocking conditions are optimized.
- Cross-reactivity is confirmed with human OPRL1; if using other species, validate sequence homology of the immunogen region.
