Biological Background: OPN5 Function and Localization
- OPN5, also known as neuropsin, GPR136, or TMEM13, is a G protein-coupled opsin receptor that functions as a photoreceptor in the retina and other tissues.
- It acts as a G protein-coupled photoreceptor that selectively activates G(i) protein in response to light. Activation involves light-induced isomerization of the opsin-bound cis-retinal to all-trans-retinal, triggering a phototransduction cascade. The protein is bistable, absorbing UVA (380 nm) and blue (470 nm) light for photoreversible regeneration. Related references: PMID:22043319
- Required for light response in the inner plexiform layer and contributes to regulation in the nerve fiber layer via phosphorylated DAT/SLC6A3 dopamine uptake modulation.
- Involved in local corneal and retinal circadian rhythm photoentrainment by modulating UVA light-induced phase-shift of the retina clock. Acts as a circadian photoreceptor in the outer ear and vibrissal pads, regulating clock-gene expression in response to violet light.
- Plays a role in postnatal retinal development by negatively regulating hyaloid vessel regression through light-dependent OPN5-SLC32A1-DRD2-VEGFR2 signaling.
- Mediates violet light-accelerated corneal wound healing, being transiently expressed after physical injury.
- Localizes to the cell membrane and is detected in the brain, retina, and neural retina-derived cell lines.
- Contains conserved features of G protein-coupled receptors, including transmembrane domains, disulfide bonds, and palmitoylation sites, and belongs to the opsin family of photoreceptor proteins.
Experimental Guidance and Technical Tips
- This antibody was generated against a synthetic peptide corresponding to amino acids 200-300 of human OPN5. Validate specificity in your target species (human, mouse, rat) by using appropriate positive and negative controls.
- For western blotting, the predicted molecular weight of OPN5 is approximately 40 kDa (UniProt: 39.7 kDa). Ensure proper electrophoresis conditions and confirm band identity with lysates from OPN5-expressing tissues such as retina or neural retina-derived cell lines.
- ELISA-based detection may require optimization of coating antigen concentration and antibody titer. Consider using recombinant OPN5 or peptide as coating antigen.
- Cross-reactivity with mouse and rat OPN5 is supported by sequence homology; however, performance in these species should be independently validated.
