Biological Background: OMA1 Function and Localization

  • Metalloendopeptidase OMA1, mitochondrial, encoded by the OMA1 gene; also known as metalloprotease-related protein 1 (MPRP-1) and designated as DAB1, YKR087C, ZMPOMA1, or 2010001O09Rik.
  • Functions as a stress-activated metalloprotease in the mitochondrial inner membrane quality control system, cleaving key substrates OPA1, UQCC3, and DELE1 to regulate mitochondrial dynamics and signaling. Related references: PMID:20038677, PMID:25605331, PMID:32132706
  • Regulates mitochondrial inner membrane fusion by cleaving OPA1 at the S1 site, generating soluble S-OPA1 that cooperates with membrane-anchored L-OPA1; under stress, excessive cleavage inhibits fusion. Related references: PMID:31922487, PMID:20038677, PMID:25275009
  • Acts as a pro-apoptotic regulator: upon BAK/BAX aggregation, OMA1-mediated OPA1 cleavage remodels cristae and facilitates cytochrome c release. Related references: PMID:25275009
  • Serves as a backup protease for PINK1 in depolarized mitochondria, cleaving PINK1 to promote its proteasomal degradation. Related references: PMID:30733118
  • Activates the integrated stress response (ISR) by processing DELE1 into S-DELE1, which translocates to the cytosol and triggers EIF2AK1/HRI kinase signaling. Related references: PMID:32132706, PMID:32132707
  • Localizes to the mitochondrial inner membrane.
  • Widely expressed with highest levels in heart, skeletal muscle, kidney, and liver.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 360–524 of human OMA1 (NP_660286.1), which corresponds to a conserved C-terminal region potentially involved in substrate recognition; consider epitope mapping if precise binding site identification is required.
  • Predicted molecular weight is approximately 60 kDa; validate in the relevant sample system with appropriate loading controls and consider mitochondrial enrichment to enhance detection.
  • Suitable for Western blot (WB) and ELISA; for other applications such as immunohistochemistry or immunofluorescence, titrate and validate under specific experimental conditions.
  • OMA1 is activated by mitochondrial stress (e.g., CCCP-induced membrane depolarization); for induction studies, pre-treat cells with stress agents to increase cleavage signal and optimize detection.
