Biological Background: OMA1 Function and Localization
- OMA1 is a mitochondrial inner membrane zinc metalloprotease that is activated by various mitochondrial stresses to cleave OPA1, UQCC3, and DELE1. Related references: PMID:20038677, PMID:25605331, PMID:32132706
- Cleavage of OPA1 at the S1 site generates soluble OPA1 (S-OPA1), which cooperates with the long form to mediate mitochondrial inner membrane fusion. - In apoptosis, OMA1 mediates OPA1 cleavage upon BAK/BAX aggregation, leading to cristae remodeling and cytochrome c release. Related references: PMID:25275009
- OMA1 activates the integrated stress response by cleaving DELE1 to produce S-DELE1, which translocates to the cytosol and activates the kinase EIF2AK1/HRI. Related references: PMID:32132706, PMID:32132707

- For Western blotting, consider using mitochondria-enriched lysates and validate detection of the ~60 kDa band corresponding to full-length OMA1; processing may produce lower molecular weight fragments under stress conditions.
- The antibody is raised against a synthetic peptide (aa 421–520) near the C-terminus; it cross-reacts with mouse and rat OMA1, enabling rodent model studies—validate specificity in your model system.
