Biological Background: Olig2 Function and Localization
- Olig2 (Oligodendrocyte transcription factor 2) is a class B/E basic helix-loop-helix (bHLH) transcription factor critical for specifying oligodendrocyte and motor neuron fates in the developing spinal cord and hindbrain.
- Cooperates with OLIG1 to establish the pMN domain of the embryonic neural tube and acts as an antagonist of V2 interneuron and NKX2-2-induced V3 interneuron development.
- Orchestrates progenitor cell fates during cortical gliogenesis and gliomagenesis; in cooperation with ZNF488, promotes oligodendrocyte differentiation.
- Activates oligodendrocyte precursor cell (OPC) formation while repressing olfactory bulb interneuron intermediate progenitor generation through transcriptional silencing of GSX2 enhancer elements.
- Localizes primarily to the nucleus but can also be detected in the cytoplasm.
- Expressed in the brain, with strong expression in oligodendrogliomas; expression in astrocytomas is weak to moderate, and highly variable in glioblastomas.
- Key post-translational modifications include ubiquitin conjugation; classified as a DNA-binding developmental protein and proto-oncogene.
