Biological Background: Olig2 Function and Localization
- Encodes a basic helix-loop-helix (bHLH) transcription factor (Class B bHLHb1 / Class E bHLHe19) essential for oligodendrocyte and motor neuron specification in the spinal cord, as well as hindbrain somatic motor neuron development.
- Cooperates with OLIG1 to establish the pMN progenitor domain of the embryonic neural tube and, together with ZNF488, drives oligodendrocyte differentiation.
- Orchestrates cortical gliogenesis and gliomagenesis by activating oligodendrocyte precursor cell (OPC) formation while repressing olfactory bulb interneuron intermediate progenitor generation through transcriptional silencing of GSX2 enhancers.
- Acts as an antagonist of V2 interneuron and NKX2-2‑induced V3 interneuron development.
- Localizes predominantly to the nucleus, with additional cytoplasmic distribution.
- Highly expressed in oligodendrogliomas; expression is weak to moderate in astrocytomas and highly variable in glioblastomas.
- Classified as a developmental protein, DNA‑binding transcription factor, and proto‑oncogene with reported ubiquitin conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 168–323, a C‑terminal region of human OLIG2; confirm epitope accessibility in your assay when using endogenous or full‑length OLIG2.
- For Western blot, a band near 32 kDa is expected; optimize loading controls and transfer conditions for reliable detection.
- For immunofluorescence and immunohistochemistry, permeabilization may be necessary due to nuclear localization; validate antibody dilution and blocking in your specific tissue or cell system.
- In ELISA, consider direct or capture formats; the antibody’s performance may depend on coating antigen conformation and detection system.
- Cross‑reactivity with mouse and rat OLIG2 makes this antibody suitable for rodent model studies; verify cross‑reactivity in your experimental setup.
