Biological Background: OLFM1 Function and Localization
- Also known as Noelin, Neuronal olfactomedin-related ER localized protein, or Olfactomedin-1; belongs to the olfactomedin family.
- Functions in the regulation of axonal growth in the CNS by inhibiting the interaction between RTN4R and LINGO1, thereby preventing RTN4R-mediated axon growth cone collapse (By similarity).
- Plays a role in the production of neural crest cells from the neural tube (By similarity).
- May be required for normal olfactory responses (By similarity).
- Subcellular localization includes secretion, synapse, endoplasmic reticulum, cell projection, axon, and perikaryon.
- Post-translational modifications: N-linked glycoprotein with disulfide bonds; exists in alternative splicing isoforms.
- Key biological processes: developmental protein involved in neurogenesis and synaptic organization.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein encompassing amino acids 90-220 of human OLFM1 (NP_055094.1). This central region may detect multiple isoforms if alternative splicing occurs outside this segment.
- For Western blotting, a band around 55 kDa is expected; validate specificity using positive (e.g., brain tissue lysate) and negative controls.
- The antibody is validated for human and mouse reactivity; cross-reactivity with other species should be tested empirically.
- For ELISA, consider pairing with a monoclonal capture antibody recognizing a distinct epitope to develop a sandwich assay system.
- Optimal working dilutions must be determined experimentally for each application and sample type.
