Biological Background: OGDH Function and Localization
- OGDH encodes the E1o component (2-oxoglutarate dehydrogenase) of the multienzyme 2-oxoglutarate dehydrogenase complex (OGDHC), also known as alpha-ketoglutarate dehydrogenase (Alpha-KGDH-E1). Related references: PMID:24495017 PMID:25210035 PMID:28435050
- Catalyzes the rate-limiting, irreversible decarboxylation of 2-oxoglutarate using thiamine diphosphate (ThDP) and transfers the succinyl intermediate to the E2 subunit (DLST), a key step of the Krebs cycle. Related references: PMID:24495017 PMID:25210035 PMID:28435050
- Primarily localizes to the mitochondrial matrix, but a fraction of the OGDHC resides in the nucleus where it supplies succinyl-CoA for histone succinylation by KAT2A. Related references: PMID:29211711

Experimental Guidance and Technical Tips
- The immunogen (aa 148–427) covers a central region of OGDH, enabling recognition of both full-length and potential cleavage products; confirm detection of the expected ~116 kDa band in relevant lysates.
- For Western blot (WB), consider loading appropriate mitochondrial-enriched or whole-cell extracts and use a 6–8% SDS-PAGE gel for optimal resolution of high-molecular-weight proteins.
- When performing IHC-P or IF/ICC, include a permeabilization step to allow antibody access to mitochondrial and nuclear compartments; validate signal specificity with appropriate controls.
- In ELISA, use the antibody as a capture or detection reagent after confirming linear range and sensitivity with recombinant OGDH protein or lysate standards.
