Biological Background: ODF2 Function and Localization

  • ODF2 (Outer dense fiber protein 2), also known as cenexin, is a major component of sperm tail outer dense fibers (ODFs) and functions as a scaffold protein localized at the distal/subdistal appendages of mother centrioles.
  • It plays a critical role in centrosome organization, being required for the recruitment of PLK1 and NIN to the centrosome and for the maintenance of normal CETN1 assembly.
  • ODF2 is essential for the formation and elastic recoil of sperm tail ODFs, thereby modulating sperm motility.
  • The protein localizes to the cytoplasm, centrosome, centriole, spindle pole, and ciliary/flagellar structures, consistent with its roles in both somatic and germ cell functions.
  • Testis-specific expression at the protein level, with high abundance in the cytoplasm of step 2 round spermatids and along the middle and principal pieces of sperm tails.
  • ODF2 undergoes alternative splicing, phosphorylation, and ubiquitination, as indicated by UniProt keywords (isopeptide bond, Ubl conjugation).

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fusion protein spanning amino acids 630–829 (C-terminal region). This region may enable detection of multiple ODF2 isoforms; verify band patterns in relevant lysates.
  • For Western blot (WB), the predicted molecular weight is ~95 kDa. Use testis or sperm protein extracts as positive controls, and validate with appropriate loading controls.
  • In immunofluorescence/ICC, ODF2 shows distinct centrosomal and centriolar localization. Suitable fixation/permeabilization protocols should be optimized to preserve these structures.
  • For ELISA, coating with recombinant protein corresponding to the immunogen region is recommended. Titrate both capture and detection components in the sample system to achieve optimal signal.
