Biological Background: ODF1 Function and Localization
- ODF1 (Outer dense fiber protein 1) is also known as Heat shock protein beta-10 (HspB10) and is a member of the heat shock protein family B member 10.
- It is a major component of the outer dense fibers (ODF) of spermatozoa, which are filamentous structures located outside the axoneme in the midpiece and principal piece of the sperm tail.
- ODFs contribute to the passive elastic structures and elastic recoil of the sperm tail, essential for sperm motility.
- The protein is specifically expressed in the testis, consistent with its role in spermatogenesis and sperm differentiation.
- Subcellularly, ODF1 localizes to the sperm flagellum, including the axoneme and centrosome, as well as to the cytoplasm and cytoskeleton.

- For Western blot (WB), consider using reduced and non-reduced sperm or testis lysates. A band near 28 kDa is expected; note that ODF1 may form higher-order structures under non-reducing conditions.
- For ELISA, the antibody can be used for capture or detection of recombinant or endogenous ODF1; however, cross-reactivity with mouse and rat orthologs may allow multi-species applications with appropriate validation.
- Given the testis-specific expression, it is advisable to include negative controls from non-testicular tissues to confirm specificity.
