Biological Background: ODC1 Function and Localization
- Ornithine decarboxylase (ODC1) is the first and rate-limiting enzyme in polyamine biosynthesis, converting ornithine to putrescine, a precursor for spermidine and spermine.
- Polyamines play essential roles in cell proliferation, DNA replication, and apoptosis, linking ODC1 activity to fundamental cellular processes.
- ODC1 is a pyridoxal phosphate-dependent decarboxylase belonging to the lyase class, and is regulated by phosphorylation and S-nitrosylation.
- Mutations in ODC1 are associated with human diseases, including hypotrichosis (hair loss), as indicated by the disease variant keyword.
- The protein has a molecular weight of approximately 51 kDa and is widely studied as a target in cancer and proliferative disorders.
Experimental Guidance and Technical Tips
- For Western blot (WB), this antibody may detect a band at approximately 51 kDa corresponding to endogenous ODC1 in human cell lysates. Optimize loading and exposure due to potentially low expression levels.
- For ELISA, consider using the immunogen peptide or recombinant ODC1 protein as coating antigen; validate antibody dilution and detection conditions in your system.
- Since the immunogen is a synthetic peptide from the N-terminus (aa 1-100), the epitope is likely in this region. Confirm reactivity with full-length ODC1 and relevant isoforms.
- Cross-reactivity with other species has not been reported; validate in the species of interest before use.
