  • OCT4, also known as Oct-3, Oct-4, or OTF-3, is a POU domain transcription factor derived from the POU5F1 gene. It is a key developmental protein with multiple isoforms.
  • The protein binds specifically to the octamer motif (5'-ATTTGCAT-3') on DNA, regulating gene expression.
  • OCT4 forms a trimeric complex with SOX2 or SOX15 on DNA, controlling the expression of critical embryonic development genes such as YES1, FGF4, UTF1, and ZFP206.
  • It is indispensable for early embryogenesis and for the maintenance of embryonic stem cell pluripotency.
  • Subcellularly, OCT4 localizes to both the cytoplasm and the nucleus, consistent with its role as a transcription factor.
  • Tissue expression is predominantly observed in the developing brain, with high levels in specific cell layers of the cortex, olfactory bulb, hippocampus, and cerebellum. Expression is low in adult tissues.
  • The protein is subject to post-translational modifications, including phosphorylation and ubiquitination (Ubl conjugation), which may regulate its activity and stability.
