Biological Background: OCT4 (POU5F1) Function and Localization
- OCT4, also known as Oct-3, Oct-4, or OTF-3, is a POU domain transcription factor derived from the POU5F1 gene. It is a key developmental protein with multiple isoforms.
- The protein binds specifically to the octamer motif (5'-ATTTGCAT-3') on DNA, regulating gene expression.
- OCT4 forms a trimeric complex with SOX2 or SOX15 on DNA, controlling the expression of critical embryonic development genes such as YES1, FGF4, UTF1, and ZFP206.
- It is indispensable for early embryogenesis and for the maintenance of embryonic stem cell pluripotency.
- Subcellularly, OCT4 localizes to both the cytoplasm and the nucleus, consistent with its role as a transcription factor.
- Tissue expression is predominantly observed in the developing brain, with high levels in specific cell layers of the cortex, olfactory bulb, hippocampus, and cerebellum. Expression is low in adult tissues.
- The protein is subject to post-translational modifications, including phosphorylation and ubiquitination (Ubl conjugation), which may regulate its activity and stability.

- The immunogen used is a synthetic peptide from amino acids 210-310 of human OCT4. As a polyclonal antibody, it may recognize multiple isoforms or related family members; validate specificity in your experimental system.
- For Western blot (WB), the predicted molecular weight is approximately 39 kDa. Optimize loading controls and antibody dilutions for your specific sample type.
- In immunohistochemistry (IHC-P), consider using human or mouse embryonic stem cells or developing brain tissues as positive controls. Antigen retrieval may be required.
