Biological Background: OCRL Function and Localization
- OCRL is an inositol polyphosphate 5-phosphatase encoded by the OCRL gene (also known as OCRL1). It catalyzes the hydrolysis of the 5-position phosphate from phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) and phosphatidylinositol-3,4,5-trisphosphate (PtdIns(3,4,5)P3), with highest activity towards PtdIns(4,5)P2 (PubMed:10764818, PubMed:15474001, PubMed:7761412).
Related references: PMID:10764818, PMID:15474001, PMID:7761412 - It also hydrolyzes inositol 1,4,5-trisphosphate and inositol 1,3,4,5-tetrakisphosphate (PubMed:15474001, PubMed:40969890, PubMed:7761412).
Related references: PMID:15474001, PMID:40969890, PMID:7761412 - OCRL regulates endosomal trafficking by modulating the pool of PtdIns(4,5)P2 associated with endosomes (PubMed:21971085).
Related references: PMID:21971085 - It is involved in primary cilium assembly (PubMed:22228094, PubMed:22543976).
Related references: PMID:22228094, PMID:22543976 - OCRL acts as a regulator of phagocytosis by hydrolyzing PtdIns(4,5)P2 to promote phagosome closure via attenuation of PI3K signaling (PubMed:22072788).
Related references: PMID:22072788 - Subcellularly, OCRL localizes to the cytoplasmic vesicle, phagosome membrane, early endosome membrane, clathrin-coated pits, primary cilium, photoreceptor outer segment, endosomes, Golgi apparatus, trans-Golgi network, and lysosomes.
- It is widely expressed in brain, skeletal muscle, heart, kidney, lung, placenta, fibroblasts, retina, and retinal pigment epithelium.
- Mutations in OCRL cause Lowe oculocerebrorenal syndrome (Dent disease 2), highlighting its role in cellular homeostasis.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–220) of human OCRL, suggesting this polyclonal antibody may detect full-length OCRL and N-terminal fragments. For Western blotting, consider using appropriate controls to confirm specificity.
- Given the predicted molecular weight of ~104 kDa, ensure adequate protein separation and transfer conditions for high-molecular-weight proteins.
- This antibody reacts with human, mouse, and rat OCRL; validate in the relevant species and tissue types (e.g., kidney, brain, or fibroblasts) to confirm performance.
- For ELISA applications, optimize coating antigen concentration and antibody dilution to achieve the desired sensitivity and signal-to-noise ratio.
- The antibody may be suitable for immunoprecipitation or immunofluorescence based on the target's subcellular localization; however, validation in these applications is recommended.
