Biological Background: OCA2 Function and Localization

  • OCA2, also known as P protein, melanocyte-specific transporter protein, or pink-eyed dilution protein homolog, is a melanosomal membrane protein encoded by the OCA2 gene.
  • Functions as a key component of the melanosome-specific anion (chloride) current, modulating melanosomal pH to ensure optimal tyrosinase activity required for melanogenesis and melanosome maturation. Related references: PMID:11310796, PMID:15262401, PMID:22234890
  • Plays a central role in the mammalian pigmentary system, acting as a major determinant of ethnic skin color variation and brown/blue eye color. Related references: PMID:15262401, PMID:18252222, PMID:7601462
  • Regulates post-translational processing of tyrosinase, the rate-limiting enzyme in melanin synthesis.
  • Localizes specifically to the melanosome membrane, consistent with its transporter function.
  • Expressed in melanocytes and retinal pigment epithelium; mutations in OCA2 are associated with oculocutaneous albinism type 2 (OCA2) and other pigmentary disorders.
  • The protein has a predicted molecular weight of approximately 93 kDa and is subject to alternative splicing and glycosylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal 180 amino acids of human OCA2; therefore, the antibody is expected to recognize epitopes within this region. For assays detecting endogenous OCA2, confirm recognition in your specific sample system.
  • For Western blotting, a specific band at approximately 93 kDa is anticipated in human and mouse melanocyte or RPE lysates. Optimize loading and detection conditions for your experimental setup.
  • In ELISA, consider using recombinant OCA2 protein or cell/tissue lysates as coating antigen; cross-reactivity with mouse OCA2 allows testing in mouse-derived samples.
  • As a polyclonal antibody, lot-to-lot consistency should be validated for critical quantitative assays.
  • Upon arrival (shipped on ice bags), aliquot and store at -20°C or -80°C to maintain stability.
