Biological Background: OAT3/SLC22A8 Function and Localization
- OAT3 (organic anion transporter 3), encoded by the SLC22A8 gene, functions as an organic anion/dicarboxylate exchanger, coupling organic anion uptake to the sodium gradient, and transports substrates such as estrone 3-sulfate and urate in exchange for dicarboxylates. Related references: PMID:14586168, PMID:15644426, PMID:15846473, PMID:15864504
- It plays a critical role in the renal and cerebral excretion of endogenous and exogenous organic anions, helping prevent systemic accumulation of potentially toxic compounds. Related references: PMID:11306713, PMID:14586168, PMID:15846473
- OAT3 transports a diverse array of substrates including prostaglandins (PGE2, PGF2α), neuroactive tryptophan metabolites (kynurenate, xanthurenate), biopterins (BH4, BH2, sepiapterin), steviol, and numerous drugs and xenobiotics such as fexofenadine, cimetidine, benzylpenicillin, acyclovir, ochratoxin A, as well as uremic toxins and statins. Related references: PMID:11907186, PMID:22108572, PMID:23832370, PMID:28534121, PMID:15644426, PMID:11669456, PMID:15846473, PMID:16455804, PMID:14675047
- Subcellularly, OAT3 is localized on the basolateral cell membrane, consistent with its role in transepithelial transport.
- Expression of OAT3 is highest in the kidney, with weaker expression in the brain, skeletal muscle, and adrenal glands. Related references: PMID:11306713, PMID:11912245, PMID:15864504
- OAT3 contributes to cortisol release in the adrenals and facilitates brain-to-blood efflux of statins and other organic anions across the blood-brain barrier. Related references: PMID:15864504
- UniProt keywords indicate that OAT3 is subject to alternative splicing and is a glycoprotein and phosphoprotein, suggesting regulation by post-translational modifications.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide within amino acids 443–542 of human OAT3, a region likely accessible in denatured and reduced samples; therefore, this antibody is suitable for Western blotting of kidney or brain tissue lysates under standard denaturing and reducing conditions.
- For ELISA applications, consider using indirect or capture formats with the peptide antigen as a positive control, and include negative controls (lacking antigen or using a non-specific peptide) to verify specificity.
- Since OAT3 is a moderate-abundance membrane protein, optimize lysis and blocking conditions to minimize nonspecific background.
- To maintain antibody integrity, follow storage recommendations; the antibody is shipped on ice bags and should be stored appropriately upon receipt.
