Biological Background: OAT3 Function and Localization

  • OAT3 (SLC22A8) is a sodium-independent organic anion/dicarboxylate exchanger located on the basolateral membrane, mediating the uptake of organic anions in exchange for dicarboxylates like α-ketoglutarate. Related references: PMID:14586168, PMID:15644426, PMID:15846473, PMID:15864504
  • It transports a wide array of endogenous substrates, including estrone-3-sulfate, urate, prostaglandins E2 and F2α, neuroactive tryptophan metabolites, and biopterins. Related references: PMID:14586168, PMID:15846473, PMID:15864504, PMID:11907186, PMID:22108572, PMID:23832370, PMID:28534121
  • OAT3 is a key player in the renal elimination and detoxification of numerous drugs (e.g., cimetidine, fexofenadine, acyclovir), toxins (e.g., ochratoxin A), and uremic solutes, helping to limit systemic exposure. Related references: PMID:11306713, PMID:11669456, PMID:15846473, PMID:16455804, PMID:14675047
  • The protein is predominantly expressed in the kidney, with lower levels in the brain (including the blood-brain barrier and choroid plexus), skeletal muscle, and adrenal glands. Related references: PMID:11306713, PMID:11912245, PMID:15864504
  • OAT3 undergoes post-translational modifications, including glycosylation and phosphorylation, which may influence its transporter activity and localization.
  • Alternative splicing of the SLC22A8 gene may generate multiple isoforms, potentially diversifying transport function.

Experimental Guidance and Technical Tips

  • This rabbit polyclonal antibody was raised against a recombinant protein corresponding to the N-terminal cytoplasmic domain (amino acids 32-123) of human OAT3. For immunofluorescence or immunocytochemistry, cells should be fixed and permeabilized to allow antibody access to the intracellular epitope. Titrate the antibody starting at 1:50–1:200 for IF/ICC.
  • For ELISA applications, the antibody can be used as a capture or detection reagent; test dilutions from 1:500 to 1:5000 against the recombinant protein or cell lysates.
  • The antibody is specific to human OAT3; cross-reactivity with mouse or rat orthologs has not been determined. Validate reactivity in the relevant species if using animal models.
  • As with any polyclonal antibody, lot-to-lot variability may occur. We recommend including positive and negative controls (e.g., OAT3-overexpressing cells versus knockdown controls) in all experiments.
