Biological Background: OAS1 Function and Localization

  • OAS1 encodes 2'-5'-oligoadenylate synthase 1, existing as p46 and p42 isoforms through alternative splicing, and is a member of the OAS family of interferon-induced proteins.
  • It acts as a critical innate immune sensor and antiviral enzyme, activated by double-stranded RNA (dsRNA) to catalyze 2'-5'-oligoadenylate (2-5A) synthesis from ATP. Related references: PMID:34581622
  • The 2-5A molecules bind to and dimerize latent RNase L, triggering its activation and leading to widespread RNA degradation, thereby inhibiting protein synthesis and blocking viral replication. Related references: PMID:34145065, PMID:34581622, PMID:40010341
  • OAS1 contributes to intercellular immune communication that limits the local dissemination of RNA viruses and exerts tumor-suppressive functions. Related references: PMID:40010341
  • C-terminal prenylation directs OAS1 to membranous replication organelles in SARS-CoV-2-infected cells, where it specifically binds conserved stem-loop structures in the viral 5'-UTR, activating RNase L and potently suppressing viral replication. Related references: PMID:34581622
  • Subcellular localization studies reveal OAS1 in the cytoplasm, mitochondria, nucleus, microsomes, and endoplasmic reticulum, with a fraction also secreted.
  • OAS1 is highly expressed in lung tissue, and its secreted form mediates antiviral effects against viruses such as VSV, HSV-2, and EMCV via RNase L-independent pathways.
  • Key structural and functional features include ATP-binding, nucleotide-binding, prenylation, and participation in host-virus interactions and innate immunity.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide within the N-terminal region (aa 1-100) of human OAS1. This polyclonal antibody may recognize both p46 and p42 isoforms, but consider epitope accessibility differences between native and denaturing conditions. Validate isoform detection in your model system.
  • For Western blot, the predicted molecular weight is approximately 46 kDa; however, alternative splicing or post-translational modifications may result in multiple bands. Include appropriate positive and negative controls to confirm specificity.
  • In ELISA, the polyclonal nature of this antibody can provide high sensitivity. Consider testing it as either capture or detection reagent, and optimize pairing with a monoclonal antibody for sandwich ELISA formats targeting the 2-5A pathway.
  • Reported cross-reactivity with mouse and rat suggests potential utility in rodent studies, but confirm reactivity and performance in the relevant sample matrix before use.
