- Nyctalopin (NYX) is a secreted proteoglycan with a predicted molecular weight of approximately 51.5 kDa (UniProt: Q9GZU5).
- The protein is a member of the small leucine-rich repeat proteoglycan (SLRP) family, characterized by leucine-rich repeat (LRR) motifs.
- NYX plays a critical role in vision by mediating light-induced depolarization of retinal ON-bipolar cells, likely acting as a scaffold for the proteins TRPM1 and GRM6.
- It is essential for the proper trafficking of TRPM1 to the dendritic tips of ON-bipolar cells, thereby facilitating signal transduction at the photoreceptor-bipolar synapse.
- Subcellularly, NYX is localized to the extracellular matrix, dendrites, and postsynaptic regions, and is secreted into the extracellular space.
- Tissue expression is prominent in the retina (inner segment of photoreceptors, outer and inner nuclear layers, ganglion cell layer) and kidney, with lower levels in brain, testis, and muscle.
