- NUTF2 (Nuclear Transport Factor 2), also designated NTF2 and placental protein 15 (PP15), is a 14 kDa protein belonging to the NTF2 family and plays a key role in nucleocytoplasmic transport.
- It specifically mediates the import of GDP-bound RAN from the cytoplasm into the nucleus, a critical step that enables RAN to function in cargo receptor-mediated transport across the nuclear envelope.
- NUTF2 directly interacts with GDP-bound RAN in the cytosol and guides it to the nuclear pore complex via associations with nucleoporins, thereby promoting nuclear translocation.
- Subcellularly, NUTF2 localizes to the cytoplasm, nuclear pore complex, and both inner and outer nuclear membranes, consistent with its shuttling function.

Experimental Guidance and Technical Tips
- The immunogen is a full-length recombinant human NUTF2 protein (aa 1-127), so the resulting polyclonal antibody is expected to recognize multiple epitopes, conferring high sensitivity.
- For Western blotting, NUTF2 has a predicted molecular weight of ~14 kDa. Small proteins may require optimized transfer conditions (e.g., lower methanol concentration or shorter transfer time) to ensure efficient membrane binding.
