Biological Background: NUP98 Function and Localization
- NUP98, also known as ADIR2, NUP96, NUP196, or Nup98-96, is a component of the nuclear pore complex (NPC) and is produced as a precursor protein that undergoes autocatalytic cleavage to generate NUP98 and NUP96, both integral to NPC function.
- It plays a critical role in NPC assembly and maintenance, and both NUP98 and NUP96 are required for bidirectional nucleocytoplasmic transport. Related references: PMID:33097660
- In cooperation with DHX9, NUP98 participates in transcription regulation and alternative splicing of specific genes, and is involved in localizing DHX9 to discrete intranuclear foci known as GLFG bodies. Related references: PMID:28221134

- For Western blotting, a band around 98 kDa (cleaved NUP98) or 198 kDa (precursor) may be observed depending on sample preparation and electrophoresis conditions; validate using nuclear extracts from human or mouse cells.
- For immunofluorescence/ICC, expect punctate nuclear rim staining characteristic of nuclear pore complexes; permeabilization is necessary for antibody access.
