Biological Background: NUP85 Function and Localization
- NUP85 (also known as Nup75, FROUNT, NPHS17) is a 75 kDa component of the nuclear pore complex (NPC), encoded by the NUP85 gene.
- It is an essential scaffolding nucleoporin required for NPC assembly and maintenance, and as part of the Nup107-160 subcomplex, it participates in RNA export and tethering of NUP96/Nup98 and NUP153 to the nucleus.
Related references: PMID:12718872 - The Nup107-160 complex, including NUP85, is required for spindle assembly during mitosis.
Related references: PMID:16807356 - NUP85 (also called FROUNT) mediates CCL2-CCR2 signaling by promoting membrane clustering of activated CCR2 and linking to the phosphatidyl-inositol 3-kinase-Rac-lammellipodium protrusion cascade, thereby regulating monocyte chemotaxis.
Related references: PMID:15995708 - It is involved in kidney development, with variants associated with nephrotic syndrome; plays a role in nephrogenesis.
Related references: PMID:30179222 - Subcellularly, NUP85 localizes to the nuclear pore complex, but also to the kinetochore, mitotic spindle, and cytoplasm, reflecting its dynamic roles in transport and cell division.
- Post-translational modifications include acetylation and phosphorylation (as per UniProt keywords).
- The gene undergoes alternative splicing, potentially producing multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 427-656) of human NUP85. This region is suitable for generating antibodies that recognize endogenous full-length protein.
- For Western blot (WB), consider using whole cell lysates or nuclear fractions from human, mouse, or rat cells, and validate specificity by signal at ~75 kDa.
- For ELISA, use recombinant NUP85 or peptide as coating antigen.
- Since NUP85 is part of large macromolecular complexes (Nup107-160), Co-IP or other interaction assays may require detergent and buffer optimization.
