Biological Background: NUP214 Function and Localization
- NUP214, also known as CAN protein or 214 kDa nucleoporin, is a large nucleoporin component of the nuclear pore complex (NPC).
- It has a critical role in nucleocytoplasmic transport, mediating the passage of macromolecules between the nucleus and cytoplasm.
Related references: PMID:9049309 PMID:31178128 - It serves as a docking site for receptor-mediated import of substrates across the NPC.
Related references: PMID:31178128 PMID:8108440 - During adenovirus infection, NUP214 is required for capsid disassembly and release of the viral genome into the nucleus (in vitro).
- Subcellularly, NUP214 is localized to the nuclear pore complex on the nuclear envelope.
- NUP214 is expressed in a tissue-specific manner, with highest levels in thymus, spleen, bone marrow, kidney, brain, and testis.
- It is subject to various post-translational modifications, including acetylation, phosphorylation, and glycosylation, and is involved in chromosomal rearrangements linked to oncogenesis.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide within amino acids 1251–1350 of human NUP214; thus, the antibody may recognize an internal epitope conserved across isoforms.
- For western blot, due to the large protein size (~214 kDa), consider using a low-percentage (e.g., 6–8%) SDS-PAGE gel and prolonged transfer times to ensure efficient blotting.
- In immunofluorescence/ICC, validate fixation and permeabilization conditions (e.g., methanol vs. Triton X-100) to achieve optimal nuclear pore staining.
- For ELISA, the peptide-based immunogen suggests this antibody may be suitable for sandwich or indirect ELISA formats; cross-reactivity with mouse and rat should be verified if working with those species.
- Always include appropriate positive and negative controls (e.g., HeLa lysates for WB) and titrate the antibody for each application.
