Biological Background: NUMA1 Function and Localization
- Nuclear Mitotic Apparatus Protein 1 (NUMA1), also known as NuMA, NMP-22, or SP-H antigen, is a large 238 kDa coiled-coil protein encoded by the NUMA1 gene. It is a major component of the nuclear matrix.
- NUMA1 plays a critical role in the formation and maintenance of spindle poles during mitosis by tethering microtubule minus ends, thereby ensuring proper chromosome alignment and segregation.
Related references: PMID:11956313 PMID:12445386 - It functions in mitotic spindle orientation and elongation through dynein-dynactin-dependent mechanisms, interacting with cortical complexes containing GPSM2 and Gα proteins, as well as phosphoinositides like PI(4,5)P2.
Related references: PMID:22327364 PMID:23027904 PMID:23921553 PMID:24371089 PMID:24996901 PMID:23870127 PMID:24109598 - NUMA1 is also required for mitotic microtubule aster assembly and anastral spindle formation, positively regulating TNKS localization to spindle poles.
Related references: PMID:11163243 PMID:11229403 PMID:12445386 PMID:25657325 PMID:16076287 - Beyond mitosis, NUMA1 is a highly abundant nuclear matrix protein that occupies the majority of the nuclear volume, suggesting a structural role. It is essential for epidermal differentiation and hair follicle morphogenesis.
- Subcellular localization includes the nucleus, nucleoplasm, nuclear matrix, spindle poles, centrosomes, cell cortex, and lateral cell membrane, reflecting its dynamic roles in both interphase and mitosis.
- Post-translational modifications include acetylation, ADP-ribosylation, phosphorylation, and ubiquitination, as annotated in UniProt (isopeptide bond, Ubl conjugation, glycoprotein, lipoprotein), which may regulate its functions.
Experimental Guidance and Technical Tips
- Given the large molecular weight of NUMA1 (approximately 238 kDa), for Western blotting, consider using a low-percentage polyacrylamide gel (e.g., 6–8%) and extended transfer times to ensure efficient transfer.
- The immunogen corresponds to the N-terminal region (amino acids 1–309) of human NUMA1; verify that this region is not occluded by post-translational modifications or binding partners in your samples.
- For immunofluorescence (IF/ICC), NUMA1 displays nuclear localization in interphase cells and spindle pole localization in mitotic cells; optimize fixation and permeabilization conditions to preserve antigenicity.
- In immunohistochemistry (IHC), antigen retrieval methods may be necessary for formalin-fixed paraffin-embedded tissues; validate staining patterns against known subcellular localizations.
- As a polyclonal antibody, lot-to-lot variability can occur; include appropriate positive and negative controls in each experiment.
