Biological Background: NUDT5 Function and Localization

  • NUDT5 is a dual-function enzyme: in the absence of diphosphate, it acts as an ADP-sugar pyrophosphatase, hydrolyzing various modified nucleoside diphosphates; in the presence of diphosphate, it catalyzes ATP synthesis. Related references: PMID:27257257

  • For Western blot, a starting dilution of 1:1000 is commonly used for monoclonal antibodies, but optimization is recommended using a dilution series.
  • In immunofluorescence/ICC, permeabilization with Triton X-100 or similar detergent is required to access nuclear NUDT5; methanol fixation may also help preserve antigenicity.
