Biological Background: NUDT5 Function and Localization
- NUDT5 is a dual-function enzyme: in the absence of diphosphate, it acts as an ADP-sugar pyrophosphatase, hydrolyzing various modified nucleoside diphosphates; in the presence of diphosphate, it catalyzes ATP synthesis. Related references: PMID:27257257
- As an ADP-sugar pyrophosphatase, NUDT5 hydrolyzes substrates such as ADP-ribose, ADP-mannose, ADP-glucose, 8-oxo-GDP, and 8-oxo-dGDP with similar efficiencies. Related references: PMID:10567213, PMID:10722730, PMID:17052728

- Since the immunogen corresponds to the N-terminal region (aa 1–100), the antibody is expected to recognize full-length NUDT5 and potentially any isoforms containing this region; always validate in your cell or tissue system.
- For Western blot, a starting dilution of 1:1000 is commonly used for monoclonal antibodies, but optimization is recommended using a dilution series.
- In immunofluorescence/ICC, permeabilization with Triton X-100 or similar detergent is required to access nuclear NUDT5; methanol fixation may also help preserve antigenicity.
- Pre-adsorption with the immunizing peptide can serve as a specificity control in both WB and IF/ICC.
