Biological Background: NUDT16 Function and Localization
- NUDT16 is a multifunctional enzyme, annotated as U8 snoRNA-decapping enzyme, and also known as IDP phosphatase (IDPase), nudix hydrolase 16, and U8 snoRNA-binding protein H29K.
- In its role as an RNA decapping enzyme, NUDT16 catalyzes the metal-dependent cleavage of the cap structure of U8 snoRNA and mRNAs, essential for rRNA maturation and mRNA turnover. Related references: PMID:20385596, PMID:26121039
- The enzyme also functions as a nucleotide sanitizer, hydrolyzing non-canonical purine nucleotides (IDP, dITP, GDP, dGDP, XDP, ITP, dITP) to their monophosphate forms, with substrate preference IDP > dIDP >> GDP = dGDP > XDP = ITP = dITP, thus preventing their incorporation into nucleic acids. - NUDT16 localizes to multiple compartments including the nucleus, nucleoplasm, nucleolus, and cytoplasm, reflecting its diverse functions in RNA processing and nucleotide metabolism.

Experimental Guidance and Technical Tips
- The immunogen targets amino acids 60–130 within the Nudix domain of human NUDT16. This region shows high conservation between human and mouse, consistent with the cross-reactivity data.
- For Western blot detection, use moderate amounts of whole cell lysate from human or mouse cell lines (e.g., 20–30 µg), and probe with standard chemiluminescent protocols. A band near 21 kDa is expected.
- In ELISA applications, the antibody may serve as a capture or detection reagent. Optimization of coating concentration and paired detection system is recommended.
- Because NUDT16 is a low-abundance protein in some cell types, enrichment of nuclear fractions or use of more sensitive detection reagents may improve signal.
- For functional studies, note that NUDT16 binds RNA and metal ions; the antibody likely recognizes total NUDT16 irrespective of its binding state.
