Biological Background: NUDT15 Function and Localization
- NUDT15, also known as MTH2 or nudix hydrolase 15, belongs to the Nudix (nucleoside diphosphate-linked moiety X) hydrolase superfamily and functions as a nucleotide triphosphate diphosphatase.
- Catalyzes the hydrolysis of nucleoside triphosphates including dGTP, dTTP, dCTP, their oxidized forms (e.g., 8-oxo-dGTP), and thiopurine prodrug derivatives 6-thio-dGTP and 6-thio-GTP. Related references: PMID:26238318
- May also hydrolyze some nucleoside diphosphate derivatives. Related references: PMID:22556419 PMID:26238318
- Hydrolyzes oxidized nucleoside triphosphates such as 8-oxo-dGTP in vitro, though the physiological relevance as a sanitizing enzyme remains unclear. Related references: PMID:22556419 PMID:26238318
- Through hydrolysis of thioguanosine triphosphates, may participate in the catabolism of thiopurine drugs. Related references: PMID:25108385 PMID:26238318
- May have a role in DNA synthesis and cell cycle progression by stabilizing PCNA. Related references: PMID:19419956
- Exhibits decapping activity towards dpCoA-capped RNAs in vitro.
- Catalyzes the hydrolysis of isoprene pyrophosphates such as geranyl diphosphate and farnesyl diphosphate in vitro, potentially contributing to the regulation of isoprenoid synthesis and protein isoprenylation. Related references: PMID:38944687
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-164 of human NUDT15, covering the full-length protein. This antibody is expected to recognize the native, full-length target in appropriate sample types.
- For Western blotting, a band near 19 kDa is expected. Consider routine validation steps, such as positive and negative controls, to confirm specificity in your lysate of interest.
- For immunohistochemistry, antigen retrieval optimization (e.g., heat-induced epitope retrieval in citrate or EDTA buffer) may improve staining results. Titrate antibody concentration to achieve optimal signal-to-background ratio.
- This polyclonal antibody cross-reacts with mouse and rat NUDT15, making it suitable for studies in rodent models.
- As with any polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative assays.
