Biological Background: NUDEL/NDEL1 Function and Localization
- NUDEL/NDEL1 is a 38 kDa coiled-coil protein critical for microtubule organization and dynein regulation; encoded by NDEL1 (also known as MITAP1).
- It anchors microtubules at the centrosome by targeting KATNA1 and enhances dynein-mediated transport; essential for Golgi integrity, vesicle trafficking, and nuclear-centrosome coupling.
- Crucial for neuronal migration during corticogenesis and, together with DISC1, promotes neurite outgrowth.
- Displays cell-type-specific roles in mitosis: required in some cells but dispensable in cortical neural progenitors (replaced by NDE1).
- Promotes neurofilament assembly and regulates lysosome distribution and osteoclast ruffled border formation (by similarity).
- Serves as a RAB9A/B effector for retrograde endosome-to-Golgi transport. Related references: PMID:34793709
- Subcellularly located at the centrosome, mitotic spindle, kinetochore, and cytoplasm; strongly expressed in brain, heart, kidney, liver, lung, pancreas, placenta, and skeletal muscle.

- The immunogen (aa 1–100) is highly conserved across human, mouse, and rat; cross-reactivity with these species is indicated.
- For Western blot, optimize primary dilution (typical range for monoclonals: 1:500–1:2000); expect a band near 38 kDa, with possible additional bands due to alternative splicing or PTMs.
- For ELISA, titrate the antibody as detection or capture reagent in indirect or sandwich formats.
- For centrosome or spindle localization studies, use appropriate cell synchronization and fixation protocols.
- Store at –20°C upon receipt; minimize freeze-thaw cycles.
