Biological Background: THOC1/p84 Function and Localization
- THOC1 couples mRNA transcription, processing, and nuclear export by associating with spliced mRNA and promoting NXF1-mediated export. The THOC1-2-3 core complex directly binds NXF1-NXT1 and stimulates DDX39B ATPase activity.

- THOC1 regulates transcriptional elongation of specific genes, contributing to gene expression control.
Related references: PMID:22144908
- It helps maintain genome stability by preventing co-transcriptional R-loop formation.
- Subcellularly, THOC1 localizes to nuclear speckles, nucleoplasm, nuclear matrix, and the cytoplasm.
- THOC1 participates in an apoptotic pathway involving caspase-6 activation, upregulation of BAK1/BCL2L1, and NF-kappa-B activation, independent of p53, and imposes a G2/M checkpoint; apoptosis is inhibited by RB1 binding.
- THOC1 is ubiquitously expressed, with very low levels in normal breast epithelium but high expression in breast tumors, correlating with aggressive phenotype, tumor size, and metastasis.
- Mutations in THOC1 are linked to autosomal dominant deafness DFNA86, possibly through hair cell dysfunction.
- The protein is subject to acetylation, phosphorylation, and ubiquitin-like modifications.
Experimental Guidance and Technical Tips
- The immunogen encompasses the C-terminal region (aa 428-657) of human THOC1, which may confer target specificity; consider confirming epitope recognition under your experimental conditions.
- This polyclonal antibody is suitable for WB, IHC-P, IF/ICC, and ELISA; titrating the antibody and optimizing protocol parameters is recommended for each assay.
- The antibody cross-reacts with mouse and rat THOC1, allowing multispecies studies; validate cross-reactivity in your specific models.
- Expected molecular weight is approximately 76 kDa; use appropriate molecular weight markers and verify the band in your sample type.
- For immunohistochemistry, consider antigen retrieval and use tissue sections known to overexpress THOC1 (e.g., breast carcinoma) as positive controls.
