Biological Background: NUBP1 Function and Localization
- NUBP1, also known as nucleotide-binding protein 1 (NBP1) or CIAO5, is a cytosolic Fe-S cluster assembly factor encoded by the NUBP1 gene in humans. Alternative splicing generates multiple isoforms.
- It is a key component of the cytosolic iron-sulfur (Fe/S) protein assembly (CIA) machinery, which is required for the maturation of extramitochondrial Fe-S proteins. NUBP1 forms a heterotetramer with NUBP2 that functions as a scaffold for de novo Fe-S cluster assembly and transfer to target apoproteins.
Related references:
PMID:18573874 - NUBP1 has been implicated in the regulation of centrosome duplication and negatively regulates cilium formation and structure, linking Fe-S cluster biogenesis to cell cycle and cytoskeletal organization.
- Subcellularly, NUBP1 is localized to the cytoplasm and nucleus, and it associates with various cytoskeletal structures, including the centrosome, centriole, basal body, cilium axoneme, and microtubule organizing center.
- The protein contains a 4Fe-4S cluster, an ATP-binding domain, and undergoes post-translational modifications such as acetylation and phosphorylation, reflecting its dynamic regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 201-320 of human NUBP1, suggesting that the antibody likely recognizes an internal epitope. Consider this when designing blocking peptides or alternative detection strategies.
- For Western blot (WB), expect a band at approximately 35 kDa (consistent with the predicted molecular weight of 34.5 kDa). Validate specificity using positive and negative control lysates.
- For immunohistochemistry (IHC-P), validate the antibody in relevant human, mouse, and rat tissue sections, considering the reported cross-reactivity. Optimize antigen retrieval and antibody dilution based on your specific experimental conditions.
- In ELISA applications, the antibody may be used for direct detection or as a capture reagent; empirical optimization of coating and detection conditions is recommended.
