Biological Background: NUAK2 Function and Localization
- NUAK2 (NUAK family SNF1-like kinase 2), also known as SNARK or Omphalocele kinase 2, is a 69 kDa serine/threonine-protein kinase belonging to the AMPK-related kinase family.
- Acts as a stress-activated kinase that promotes tolerance to glucose starvation, enabling cellular survival under metabolic stress.
- Induces cell-cell detachment by facilitating F-actin to G-actin conversion, and promotes cell motility and invasiveness in tumor cells upon CD95/TNF-alpha stimulation via NF-kappa-B signaling.
- Protects cells from CD95-mediated apoptosis, suggesting a role in survival signaling.
- Phosphorylates LATS1 and LATS2, key kinases in the Hippo signaling pathway, thereby regulating downstream YAP1 nuclear localization.
- Critical for neural tube closure during embryonic development through LATS2 phosphorylation and subsequent YAP1 regulation. Related references: PMID:32845958
- Post-translationally modified by phosphorylation and acetylation, as indicated by proteomics identification and direct protein sequencing evidence.

- The immunogen used corresponds to the C-terminal region (amino acids 479-628) of human NUAK2, which is less conserved among related kinases. This design may enhance specificity; however, cross-reactivity should always be validated in your experimental system.
- For IF/ICC applications, optimize fixation and permeabilization conditions based on your cell type. Consider using human cell lines known to express NUAK2 (e.g., tumor cells with active CD95/TNF-alpha signaling) as positive controls.
- Due to confirmed human reactivity only, ensure that your sample species is compatible. If other species are needed, request a BLAST analysis or homology test in advance.
