Biological Background: NUAK1 Function and Localization
- NUAK1, also known as ARK5 (AMPK-related protein kinase 5) or omphalocele kinase 1, is a serine/threonine-protein kinase belonging to the NUAK family of AMPK-related kinases.
- Controls cell adhesion by phosphorylating the myosin phosphatase target subunit PPP1R12A, which then binds 14-3-3 proteins, leading to decreased myosin light chain dephosphorylation and altered actomyosin contractility.
- Regulates cellular senescence and ploidy through phosphorylation of LATS1 at Ser-464, affecting its stability and consequently the Hippo signaling pathway.
- Participates in the DNA damage response by phosphorylating p53/TP53 at Ser-15 and Ser-392, and is recruited to the CDKN1A/WAF1 promoter to enhance p53-dependent transcription.
- Mediates UV radiation-induced DNA damage response: in complex with STK11, phosphorylates CDKN1A, promoting its degradation to enable efficient DNA repair. Related references: PMID:25329316
