Biological Background: NT5E/CD73 Function and Localization
- Encoded by the NT5E gene, the protein is also known as 5'-nucleotidase, ecto-5'-nucleotidase, or CD73, with aliases including NT, eN, NT5, NTE, eNT, E5NT, and CALJA.
- Catalyzes the hydrolysis of nucleotide monophosphates, releasing inorganic phosphate and the corresponding nucleoside, with AMP as the preferred substrate. Related references: PMID:21933152, PMID:22997138, PMID:23142347
- Shows a preference for ribonucleotide monophosphates over deoxyribose forms, and can also hydrolyze IMP, UMP, GMP, CMP, dAMP, dCMP, dTMP, NAD, and NMN. Related references: PMID:34403084
- Localizes to the cell membrane and is attached via a GPI anchor, classifying it as an ectoenzyme.
- Contains structural features including a signal peptide, disulfide bonds, and a zinc-binding site, and undergoes glycosylation.
- Exhibits alternative splicing, resulting in multiple isoforms.
- Associated with disease variants, as indicated by its inclusion in curated disease databases, though specific mutations are not detailed here.
Experimental Guidance and Technical Tips
- The immunogen covers a large extracellular portion (amino acids 27–547), which may enhance recognition of native conformations in applications like IF-P and IHC-P.
- For western blotting, the predicted molecular weight ranges from 57 to 63 kDa; however, due to glycosylation, the observed band may migrate at a higher molecular weight. Validate with appropriate controls in your experimental system.
- In immunohistochemistry and immunofluorescence, cell membrane staining is expected. Antigen retrieval may be necessary for formalin-fixed paraffin-embedded tissues.
- Cross-reactivity with mouse and rat is reported, making it suitable for studies across these species.
- As with any antibody, individual optimization of dilution factors and conditions is recommended for each assay.
