Biological Background: NSUN3 Function and Localization

  • NSUN3 is a member of the NOL1/NOP2/Sun domain family of RNA methyltransferases and is also referred to as NOL1/NOP2/Sun domain family member 3.
  • NSUN3 catalyzes the methylation of cytosine-34 in mitochondrial tRNA-Met to 5-methylcytosine (m5C), which is subsequently oxidized to 5-formylcytosine (f5c), enabling the anticodon to decode both AUG and AUA codons, thus expanding the codon recognition capacity of mitochondrial translation. • The protein is localized to the mitochondrial matrix, consistent with its role in decorating organellar tRNAs.
  • Mutations in NSUN3 lead to combined oxidative phosphorylation deficiency 48 (COXPD48), a primary mitochondrial disease, underscoring its essential role in mitochondrial gene expression.
  • As an S-adenosyl-L-methionine-dependent methyltransferase, NSUN3 specifically binds mitochondrial tRNA-Met and has been catalogued in large-scale proteomics efforts.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–120) of human NSUN3; consider this when designing blocking peptides or competition assays.
  • Based on the predicted molecular weight (38 kDa), a band near 38 kDa is expected in western blotting; validate with appropriate positive controls (e.g., mitochondrial-enriched fractions from human, mouse, or rat cell lines).
  • For ELISA, ensure the coating antigen preserves the immunogen epitopes; a direct ELISA with the recombinant protein may be used for titer determination.
  • Cross-reactivity is reported for human, mouse, and rat; however, validate in your specific sample system as sequence homology in other regions may affect binding.
