Biological Background: NSUN2 Function and Localization
- NSUN2, also known as MISU, MRT5, SAKI, or TRM4, is an RNA cytosine C(5)-methyltransferase that catalyzes the formation of 5-methylcytosine (m5C) in various RNA species, including tRNAs, mRNAs, and long non-coding RNAs. Related references: PMID:17071714 PMID:22995836 PMID:31199786
- It methylates specific cytosine residues in tRNA precursors: positions 34 and 48 of intron-containing tRNA(Leu)(CAA) and positions 48–50 of tRNA(Gly)(GCC), a modification essential for tRNA stability and generation of tRNA-derived fragments. Experimental Guidance and Technical Tips
- For Western blotting, consider loading 20–50 µg of total protein lysate; the endogenous NSUN2 band appears at approximately 86 kDa. Optimize blocking and primary antibody dilution for your sample.
- In immunohistochemistry (paraffin sections), heat-induced antigen retrieval (e.g., citrate buffer, pH 6.0) is recommended; titrate the antibody to achieve specific staining.
- For immunofluorescence, test fixation with 4% paraformaldehyde and permeabilization with 0.1% Triton X-100. Include appropriate controls to confirm localization.
