Biological Background: NRD1 (Nardilysin) Function and Localization
- NRD1, also known as nardilysin or N-arginine dibasic convertase (NRD-C), is a metalloprotease that cleaves peptide substrates at the N-terminus of arginine residues in dibasic pairs.
- It acts as a critical activator of BACE1- and ADAM17-mediated pro-neuregulin ectodomain shedding, which is involved in positive regulation of axonal maturation and myelination.
- NRD1 is required for proper functioning of the 2-oxoglutarate dehydrogenase (OGDH) complex, a key enzyme in the citric acid cycle.
- The protein localizes to mitochondria and to cell projections such as dendrites, consistent with roles in energy metabolism and neuronal function.
- It is predominantly expressed in adult heart, skeletal muscle, and testis, with lower levels in other tissues.
- NRD1 is classified as a zinc-dependent metalloprotease and undergoes alternative splicing and phosphorylation.
- The UniProt record includes keywords such as Hydrolase, Metal-binding, Metalloprotease, Mitochondrion, Phosphoprotein, Protease, Signal, and Zinc.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 160–400 of human NRD1; verify that the antibody recognizes your isoform of interest.
- For Western blot, a band near 132 kDa is expected; high-molecular-weight proteins may require extended transfer times or low-percentage gels.
- The antibody is reported to cross-react with human, mouse, and rat; for other species, check sequence homology before use.
- When establishing an ELISA, consider pairing this antibody with an appropriate capture or detection reagent and perform titration experiments.
- Non-specific binding can be minimized by optimizing blocking conditions and antibody dilution.
