Biological Background: NR2C2 Function and Localization

  • NR2C2, also known as TR4 or TAK1, is an orphan nuclear receptor that functions as a transcriptional repressor or activator. It belongs to the nuclear receptor subfamily 2 group C.
  • It represses signaling pathways of retinoic acid receptor, retinoid X, vitamin D3 receptor, thyroid hormone receptor, and estrogen receptor, and acts as an antagonist of PPARA-mediated transactivation.
  • Together with NR2C1, it forms the DRED complex that represses embryonic and fetal globin transcription, including GATA1, by binding to direct repeat hormone response elements (AGGTCA half-sites).
  • NR2C2 plays a fundamental role in early embryonic development and embryonic stem cell maintenance, and is required for normal spermatogenesis and cerebellum development.
  • It localizes to the nucleus and binds DNA via its zinc-finger domains; it is subject to post-translational modifications including acetylation, phosphorylation, and ubiquitination.
  • The protein activates the transcriptional activity of LHCG and is important for neurodevelopmentally regulated behavior (by similarity).
