Biological Background: NQO1 Function and Localization

  • NQO1 (NAD(P)H dehydrogenase [quinone] 1) is a flavin-containing quinone reductase also known as DT-diaphorase (DTD), menadione reductase, and quinone reductase 1 (QR1). Encoded by the NQO1 gene (also DIA4, NMOR1) with alternative splicing isoforms.
  • Catalyzes two-electron reduction of quinones to hydroquinones using NADH or NADPH, bypassing semiquinone formation and reactive oxygen species generation (By similarity) (PubMed:8999809, PubMed:9271353). Related references: PMID:8999809 PMID:9271353
  • Cellular redox regulation: Reduces components of the plasma membrane redox system such as coenzyme Q and vitamin quinones, generating antioxidant hydroquinones and acting as a superoxide scavenger (PubMed:15102952, PubMed:8999809, PubMed:9271353). Related references: PMID:15102952 PMID:8999809 PMID:9271353
  • Can activate quinones and derivatives to generate redox-reactive hydroquinones with DNA cross-linking and potential antitumor activity (PubMed:8999809). Related references: PMID:8999809
  • Interacts with tumor suppressors TP53 and TP73 in a NADH-dependent manner, inhibiting their ubiquitin-independent degradation by the 20S proteasome (PubMed:15687255, PubMed:28291250). Related references: PMID:15687255 PMID:28291250
  • Subcellular localization: Cytoplasm, cytosol.
  • Keywords: FAD, flavoprotein, isopeptide bond, phosphoprotein, Ubl conjugation.

Experimental Guidance and Technical Tips

  • Immunogen design: Raised against recombinant full-length human NQO1 (aa 1-274). High conservation supports cross-reactivity with mouse and rat, as indicated.
  • Western blotting: Expect a band at approximately 31 kDa. For specificity verification, consider using NQO1 knockout cell lysates as a negative control.
  • Immunohistochemistry (IHC-P): Antigen retrieval (e.g., heat-induced epitope retrieval) is recommended for paraffin-embedded tissues. Optimize for your sample type.
  • Immunofluorescence (IF-P): NQO1 localizes to the cytoplasm; diffuse staining pattern is expected. Validate fixation and permeabilization conditions.
  • ELISA: Suitable for capture or detection; pair with a compatible secondary antibody and optimize concentrations.
